Enzymatic quantitation of pyridoxal-5-phosphat
Vitamin B6
The new vitamin B6 enzymatic assay allows for rapid and batch-wise detection of potential vitamin B6 deficiency or overdose.
Enzymatic Assay
The assay is based on a three reagent protocol employing a PLP dependent enzyme and a following colour reaction quantified in a photometer. The basic version is run in microtiter plates either by hand or pipetting robots without any washing steps.
- Measuring range: 9 – 250 nmol/l (2.2 – 62 ug/l)
- Highly comparable results to HPLC and radioenzymatic assay
- Time to result: 45 minutes
- Sample pretreatment: 1:40 sample dilution – no precipitation; no derivatisation
- Endpoint measurement at 546 nm (520 – 595 nm possible)
- Easy to adapt on liquid handling robots, no washing steps required.
- Possible automatisation on flexible Clin Chem Analyzer systems (Kone, Pentra, Mira).

Vitamin B6
Vitamin B6 is a water soluble compound found in many different foods, the biological active form, Pyridoxal 5’ phosphate (PLP) is essential for the function of more than 100 enzymes as a co-enzyme. Therefore signs of deficiency are manifold and severe states of deficiencies are seldom and mostly associated with deficiency of vitamin B12 and folate.
Large surveys in the US (NHANES) showed that even in sections of population using regularly nutritional supplements a significant part shows low plasma PLP concentrations.
Low vitamin B6 status is associated with a large amount of very different diseases including cardiovascular disease, cancer, cognitive function, premenstrual syndrome etc.
Identified risk groups are individuals with impaired renal function, autoimmune disorders including rheumatoid arthritis, malabsorbtive diseases like celiac disease, crohn’s disease and ulcerative colitis and people with alcohol dependence. Furthermore, sever interactions with medications are known for people using Cycloserine (tuberculosis antibiotic), antiepileptic medications like valproic acid and carbamacepin, and Theophylline used for treatment of asthmatic diseases.
