Oxidized DNA ELISA
Extensive experimental evidence shows that oxidative damage permanently occurs to lipids of cellular membranes, proteins, and DNA. In nuclear and mitochondrial DNA, 8-hydroxy-2' -deoxyguanosine (8-OHdG) or 8-oxo-7,8-dihydro-2' -deoxyguanosine (8-oxodG) is one of the predominant forms of free radical-induced oxidative lesions, and is therefore widely used as a biomarker for oxidative stress and carcinogenesis. Studies show that 8-OHdG is a good biomarker for risk assessment of various cancers and degenerative diseases.

Oxidative Stress
Normal metabolism of oxygen produces Reactive Oxygen Species (ROS), highly reactive molecules that can damage DNA, RNA, lipids and proteins. Whilst cells are normally able to defend themselves against such damage with a range of anti-oxidants and efficient DNA repair mechanisms increased levels of ROS lead to oxidative stress. In healthy people oxidants, anti-oxidants and repair mechanisms are balanced preventing continuous damage to genetic material.
Oxidized DNA ELISA
The Oxidized DNA ELISA from BÜHLMANN allows a direct and quantitative determination of 8-OHdG in human urine and serum. The assay is delivered as a complete kit including coated microtiter plate, calibrator and internal controls. Urine samples will be diluted 1:10 before being analysed in the assay, serum can be analysed after filtration (refer to the serum application published below).
- The BÜHLMANN oxidized DNA ELISA (EK-ODG) contains all reagents needed to run the ELISA including internal controls.
- The ELISA is a competitive assay based on a specific monoclonal antibody (86-4-2) developed by BÜHLMANN Laboratories AG.
- Except of cross-reactivity to oxidized RNA, 8-OHG (20%) and 8-OHGua, the oxidized base (17%), no other cross-reactivity was detected. Therefore, the BÜHLMANN ELISA is suitable to specifically detect the complete oxidative status of the genetic material of the organism.
- Urine is the sample of choice and can be used without extraction or pre-treatment. The sample will be used in the ELISA by a 1:10 dilution with Incubation Buffer.
- Serum can be analysed after filtration.
- The BÜHLMANN oxidized DNA ELISA correlates well with the “New 8-OHdG Check” from the Japan Institute for the Control of Aging (JaICA) which is the established ELISA on the market and used in several studies (see Download - Technical Information).
- No cross-reactivity to urea.
- The ELISA is registered for Research use only (RUO).
