Like the Flow-CAST^®, our new product Flow2 CAST^® is intended for the determination of basophil activation in patients blood samples as the test is performed on whole blood samples and the new marker combination CCR3 and CD63 combines the simple gating properties for basophil detection by CCR3 and the solid application of the proven activation marker CD63 in one new assay.

CCR3 as Basophil Marker
CCR3 is an eotaxin receptor and is constitutively expressed on basophils. CCR3 is detected with a PE-flourescence labelled monoclonal Ab. With the introduction of this development, the gating of basophil cells is significantly easier to accomplish now with the new Flow2 CAST^®, and contamination of basophils with monocytes can not occur anymore.

CD63 as Activation Marker
When basophil cells are stimulated in vitro by incubation with allergens, resulting CD63 expression on the cell surface is detected with a monoclonal antibody that is FITC-fluorescence labeled. CD63 is a reliable and the most published basophil activation marker for allergy detection.

The assay procedure times are reduced to one hour, thus building the necessary step for routine laboratories to now use this technology without the elaborate procedures that were necessary in the past.
Clinical validations already showed that the excellent quality in terms of sensitivity and specificity to a variety of allergens including drug hypersensitivities, is at least at the level of the Flow-CAST^®.
